Dissecting Molecular Heterogeneity and Clonal Evolution in Acute Leukemias Through Single-Cell Genomics

Bulk sequencing can show which mutations are present in a leukemia. It can’t show which ones occur in the same cells, the order they arose in, or how a genotype relates to a cell’s lineage.

In this 20-minute webinar from World Single Cell Day, Dr. Ilaria Iacobucci presents three studies built on single-cell DNA and protein sequencing with the Tapestri® platform. She is a principal scientist in the Department of Pathology at St. Jude Children’s Research Hospital. The studies cover engineered acute erythroid leukemia models, B- and T-cell acute lymphoblastic leukemia, and a newly identified subtype of lineage ambiguous leukemia.

Topics include:

  • Resolving clonal architecture in CRISPR-engineered acute erythroid leukemia models, including a pipeline for large CRISPR-induced indels
  • Tracking how clones gained and selected mutations across serial transplantation
  • Separating early lineage-defining alterations from later signaling mutations in B-ALL
  • Detecting subclonal RAS mutations that bulk whole genome sequencing missed
  • Mapping clone-specific copy number losses in genes such as IKZF1, PAX5 and CDKN2A/B with custom amplicon designs
  • Linking genetic clones to lineage phenotypes in a lineage ambiguous leukemia subtype using a 145-gene panel and a 45-antibody protein panel

This session is for leukemia researchers and translational teams who want to look past which mutations are present and see how clones are built and how they evolve.
